ABSTRACT

Aims: This study aimed to investigate the levels of efficacy of anthelmintics that were most commonly used in sheep farms in Nziih locality. Study Design: Longitudinal prospective study. Place and Duration of Study: The study was carried out in Nziih locality in August 2019 and September 2020. Methodology: Resistance against Albendazole (12.5 mg/kg) and Ivermectin (0.2 mg/kg) was evaluated using faecal egg count reduction test in naturally occurring gastrointestinal strongyles of sheep in Nziih. A total of 300 young animals were randomly selected from 5 farms in 2019 and in 2020. Faecal samples were collected from each animal on the first day of treatment (D0) and collected again 14 days later (D14). Faecal Egg Counts were performed using the Willis and Mac Master techniques and resistance was declared when faecal egg count reduction (FECR) was less than 95 % and the lower limit of confidence interval (LCI) less than 90%. Pooled faecal samples for each treatment group were set for D0 and D14 to identify resistant strongyles genera by calculating persistent larval rate (PLR). Results: Resistance against Ivermectin was detected while there was none against Albendazole. Moreover, it was declared only in 2 farms out of the 5 selected. From 2019 to 2020, the status of resistance has shifted from suspicion to resistance for ivermectin and from absence of resistance to suspicion to Albendazole. Resistance was recorded in 2 sites: Pastorale1 (FECR= 85 %, LCI= 42.61 %) and Pastorale 3 (FECR=90.5 %, LCI= 72.26 %) in large flock herds and in farms where sheep were dewormed at least 4 times per year. Trichostrongylus were more persistent than Haemonchus and Oesophagostomum/Chabertia with PLR of 21.22 %, 13.71 %, and 7.40 % respectively. Conclusion: Anthelmintics must be used correctly in conjunction with farmer education in Nziih.

ABSTRACT

Abstract This study evaluated the economic impact of gastrointestinal nematode (GIN) infection in Morada Nova lambs under different parasite chemical control conditions. For this, 246 lambs, in the rainy and dry season, were randomized into groups according to their anthelmintic treatment with levamisole: control (CT: no treatment); routine treatment (RT: treated every 42 days); and targeted selective treatment (TST: treated according to the average daily weight gain, DWG). From 63 days of age (D63) to D210, the lambs were weighed and monitored for GIN infection parameters. Spending on anthelmintics in the production system was 1.3% of the total economic result. The economic result per animal (R$ 5.00 = US$ 1.00) was higher in the RT group, amounting to US$ 6.60 in the rainy and US$ 5.69 in the dry season, due to higher DWG. Thus, RT presented economic results 14.4% and 10.9% higher than CT, and 7.2% and 1.9% higher than TST, in the rainy and dry season, respectively. However, fast development of resistance made RT unfeasible. Here, the economic impact of GIN infection on a national scale is discussed, demonstrating its importance and the impossibility of profitable and sustainable sheep production without adequate control.


Resumo Este estudo avaliou o impacto econômico da infecção por nematoides gastrintestinais (NGI), em cordeiros Morada Nova, sob diferentes condições de controle químico dos parasitas. Para isso, 246 cordeiros, na estação chuvosa e seca, foram randomizados em grupos de acordo com o tratamento com levamisol: controle (TC: sem tratamento); tratamento rotineiro (TR: tratado a cada 42 dias); e tratamento seletivo direcionado (TST: tratado de acordo com o ganho de peso médio diário, GMD). Dos 63 dias de idade (D63) ao D210, os cordeiros foram pesados ​​e monitorados quanto aos parâmetros de infecção por NGI. O gasto com anti-helmínticos no sistema produtivo foi de 1,3% do resultado econômico total. O resultado econômico por animal (R$ 5,00 = US$ 1,00) foi maior no grupo RT, totalizando US$ 6,60 na estação chuvosa e US$ 5,69 na seca, devido ao maior GMD. Assim, o RT apresentou resultados econômicos 14,4% e 10,9% superiores ao TC, e 7,2% e 1,9% superiores ao TST, no período chuvoso e seco, respectivamente. Entretanto o rápido desenvolvimento de resistência inviabiliza o TR. O impacto econômico da infecção por NGI em escala nacional são aqui discutidos, demonstrando sua importância e a impossibilidade de uma ovinocultura lucrativa e sustentável sem o controle adequado.

ABSTRACT

The objective of the study was to evaluate the antiparasitic resistance against horse nematodes in the South of Rio Grande do Sul, Brazil. The results concerning the tests of anthelmintic efficacy on horses, stored in the database of the Parasitic Diseases Study Group (GEEP) - Veterinary Faculty, at the Federal University of Pelotas (UFPel), were carried out in the laboratory from 2018 to 2019. Stool samples were received from farms with breeding of adult female and male Criollo horses naturally infected, located in municipalities in the country's southern region. The antiparasitic agents tested were Triclorfon + Fenbendazole, Closantel + Albendazole, Ivermectin + Praziquantel, Fenbendazole, Ivermectin, Doramectin, Mebendazole and Moxidectin. Techniques such as Gordon and Whitlock, Coproculture and Fecal Egg Count Reduction Test were performed. Of all the antiparasitic drugs tested, it was observed that only treatments with Ivermectin 2% showed desired values. The observed results indicate that resistance to macrocyclic lactones is usual in equine parasites in this Brazilian region, despite the results with isolated Ivermectin.(AU)


O objetivo deste estudo é avaliar a resistência antiparasitária contra nematodeos de equinos no sul do Rio Grande do Sul, Brasil. Os resultados referentes aos testes de eficácia anti-helmíntica em cavalos, armazenados no banco de dados do Grupo de Estudos de Doenças Parasitárias (GEEP) - Faculdade de Veterinária, da Universidade Federal de Pelotas (UFPel), foram realizados em laboratório, no período de 2018 a 2019. Amostras de fezes foram recebidas de fazendas com criação de cavalos Crioulos adultos fêmeas e machos naturalmente infectados, localizadas em municípios da região Sul do país. Os agentes antiparasitários testados foram triclorfon + fenbendazol, closantel + ivermectina + praziquantel, fenbendazol, ivermectina, doramectina, mebendazol e moxidectina. Técnicas como Gordon e Whitlock, coprocultura e teste de redução da contagem de ovos fecais foram realizadas. De todos os antiparasitários testados, observou-se que apenas os tratamentos com ivermectina 2% apresentaram os valores desejados. Os resultados indicam que a resistência às lactonas macrocíclicas é comum em parasitas equinos nessa região brasileira, apesar dos resultados com ivermectina isolada.(AU)

Abstract This study evaluated the resistance status of Haemonchus contortus from sheep flocks in the state of São Paulo, Brazil, through comparison between the fecal egg count reduction test (FECRT) and the larval development test (LDT). For the FECRT, 35 sheep were selected in each of five flocks and divided into groups treated with: benzimidazole, levamisole, ivermectin, monepantel and control. Feces were collected for EPG and fecal cultures. The LDT was performed using thiabendazole (TBZ), levamisole (LEV), ivermectin aglycone (IVM-A) and Zolvix (ZLV). Resistance to all drugs was detected using FECRT in 100% of the flocks, except in relation to ZLV (40% resistant and 20% suspected of resistance). LDT indicated resistance to TBZ and IVM-A in all flocks, to LEV in 80% of flocks and to ZLV in 10%. Total agreement was obtained between the two tests for TBZ and IVM (k = 1.0), while for LEV (k = 0.8) and ZLV (k = 0.9), substantial and almost perfect agreement were obtained, respectively. The concordance between the tests was significant, thus showing that it is possible to use the outcome of the LDT to predict the FECRT, and hence validating the former as a fast diagnostic test for use by sheep farmers in Brazil.


Resumo Este estudo avaliou o status de resistência de Haemonchus contortus em rebanhos ovinos do estado de São Paulo, Brasil, através de um estudo comparativo entre o teste de redução da contagem de ovos nas fezes (TRCOF) e o teste de desenvolvimento larvar (TDL). Para o TRCOF, 35 ovinos foram selecionados em cada um dos cinco rebanhos e divididos em grupos tratados com: benzimidazol, levamisol, ivermectina, monepantel e controle. Fezes foram coletadas para OPG e coproculturas. O TDL foi realizado com tiabendazol-TBZ, levamisol-LEV, ivermectina aglicona-IVM-A e Zolvix-ZLV. Resistência a todos os fármacos foi detectada por meio do TRCOF em 100% dos rebanhos, exceto para ZLV (40% resistentes e 20% suspeitos de resistência). O LDT indicou resistência ao TBZ e IVM-A em todos os rebanhos; em 80% dos rebanhos ao LEV e 10% ao ZLV. Foi obtida concordância total entre os dois testes para TBZ e IVM (k = 1,0), enquanto para LEV (k = 0,8) e ZLV (k = 0,9) foram obtidas concordância substancial e quase perfeita, respectivamente. A concordância entre os testes foi significante, sendo possível usar o resultado do LDT para prever o do TRCOF, validando-se o primeiro como um teste diagnóstico rápido para criadores de ovinos no Brasil.

ABSTRACT

Abstract The objective of this study was to provide the first report of resistance of Haemonchus contortus to monepantel in sheep in Espírito Santo. The study was conducted in a property with history of monepantel use since 2014 and register of low efficacy in studies conducted over the past few years with fecal egg count reduction test. Lambs born on the property (males and females aged approximately 100 days) were selected and after eggs per gram of feces (EPG) analysis on fecal samples, these were divided into two groups: a group treated with monepantel (2.5 mg / Kg) and a control group without anthelmintic treatment. Seven days later, the animals were euthanized to recover parasites from the gastrointestinal tract. The efficacy of the treatment was 61.35% against H. contortus, thus proving that anthelmintic resistance to monepantel was present.


Resumo O objetivo deste estudo foi fornecer o primeiro relato de resistência de Haemonchus contortus ao monepantel em ovinos no Espírito Santo. O estudo foi conduzido em uma propriedade com histórico de uso de monepantel, desde 2014, e registro de baixa eficácia em estudos conduzidos nos últimos anos, utilizando-se testes de redução de contagem de ovos por grama de fezes (OPG). Cordeiros nascidos na propriedade (machos e fêmeas com idade aproximada de 100 dias) foram selecionados e, após contagem de OPG, foram divididos em dois grupos: um grupo tratado com monepantel (2,5 mg / kg) e um grupo controle, sem tratamento com anti-helmíntico. Após 7 dias, os animais foram eutanasiados para a recuperação dos parasitos no trato gastrointestinal. O tratamento apresentou eficácia de 61,35% para H. contortus, comprovando que a resistência anti-helmíntica ao monepantel foi presente.

Abstract The aim of this study was to characterize the anthelmintic resistance (AR) of a sheep gastrointestinal nematode population, named Caucaia, from northeastern Brazil. Phenotypic tests performed were: egg hatch (EHT), larval development (LDT) and fecal egg count reduction (FECRT). Benzimidazoles (BZs) genotypic evaluation was by frequency of single nucleotide polymorphisms (SNPs) F200Y, F167Y and E198A, and for levamisole (LEV), by frequency of resistance alleles of Hco-acr-8 gene. The primers were designed specifically for Haemonchus contortus. Effective concentrations 50% (EC50) for BZs (EHT), and for macrocyclic lactones (MLs) and LEV (LDT) were 1.02 µg/mL, 1.81 ng/mL and 0.04 µg/mL, respectively. Resistance ratios for MLs and LEV were 0.91 and 3.07, respectively. FECRT efficacies of BZs, MLs, monepantel (MPTL) and LEV were 52.4; 87.0; 94.5 and 99.6%, respectively. qPCR for BZs demonstrated resistance allele frequencies of 0%, 26.24% and 69.08% for SNPs E198A, F200Y and F167Y, respectively. For LEV, 54.37% of resistance alleles were found. There was agreement between EHT, FECRT and qPCR for BZs, and agreement between LDT and qPCR for LEV. Thus, based on higher sensitivity of qPCR, and phenotypic evaluation, the Caucaia population was considered resistant to BZs, MLs, LEV and suspect for MPTL.


Resumo O objetivo deste estudo foi caracterizar a resistência anti-helmíntica (RA) da população de nematoides gastrintestinais de ovinos, denominada Caucaia, do Nordeste brasileiro. Os testes fenotípicos foram: eclosão de ovos (TEO), desenvolvimento larvar (TDL) e redução da contagem de ovos nas fezes (TRCOF). A avaliação genotípica para benzimidazóis (BZs) foi por frequência de polimorfismos de nucleotídeo único (SNPs) F200Y, F167Y e E198A; e para levamisol (LEV), pela frequência alélica para resistência no gene Hco-acr-8. Os "primers" foram específicos para Haemonchus contortus. As concentrações efetivas 50% (CE50) para BZs (TEO) e para lactonas macrocíclicas (LMs) e LEV (TDL) foram 1,02 µg/mL, 1,81 ng/mL e 0,04 µg/mL, respectivamente. Os fatores de resistência para LMs e LEV foram 0,91 e 3,07, respectivamente. As eficácias para BZs, LMs, monepantel (MPTL) e LEV no TRCOF foram 52,4; 87,0; 94,5 e 99,6%, respectivamente. A qPCR para BZs demonstrou frequências de 0%, 26,24% e 69,08% para SNPs E198A, F200Y e F167Y, respectivamente. Para LEV foram encontrados 54,37% de alelos resistentes. Houve concordância entre TEO, TRCOF e qPCR para BZs, e entre TDL e qPCR para LEV. Baseada na maior sensibilidade da qPCR e avaliação fenotípica, a população Caucaia foi considerada resistente a BZs, LMs, LEV e suspeita para MPTL.

Resumen En la Argentina, el 93% de la producción caprina en la Argentina es desarrollada por pequeños productores de agriculturas de subsistencia, quienes, por lo general, utilizan antihelmínticos sin asesoramiento profesional, lo que, probablemente, ha llevado al desarrollo de resistencia, comprometiendo seriamente la sustentabilidad del control de nematodos. El objetivo de este estudio es determinar la eficacia de la ivermectina, el albendazol y el levamisol para el tratamiento de la gastroenteritis verminosa en cuatro grupos de ocho caprinos cada uno, mediante el Test de Reducción del Conteo de Huevos (TRCH). Los resultados demostraron un 100% de reducción del número de HPG para los grupos tratados con albendazol y levamisol y un 17% de reducción para el grupo tratado con ivermectina, con respecto al grupo control, lo que permite inferir que existe resistencia a este fármaco.


Abstract Determination of the efficacy of anthelmintics used in the control of gastrointestinal nematodes of goats from Tucumán, Argentina. Almost 93% of goat production in Argentina is developed by small farmers, who generally use anthelmintics without professional advice, which has led to the development of resistance, seriously compromising the sustainability of nematode control. This study aims to determine the efficacy of ivermectin, albendazole and levamisole for the treatment of verminous gastroenteritis in four groups of eight goats each, using the Faecal Egg Count Reduction Test (FECRT). The results showed a 100% reduction in the number of EPG for the groups treated with albendazole and levamisole and a 17% reduction for the group treated with ivermectin, compared to the control group, which allows us to infer that there is resistance to this drug.

Abstract The aim here is to present data on the efficacy of anthelmintics in sheep flocks in Rio de Janeiro, Brazil, and to discuss the interpretation of the fecal egg count reduction test (FECRT) for each nematode genus. Fecal eggs counts and pre- and post-treatment coprocultures were performed, the former to evaluate the efficacy of and the latter to determine the overall parasite prevalence. An additional efficacy test was performed at Farm # 1 a year after the initial test. Severe anthelmintic resistance was found for the flocks, with no FECRT sensitivity at any of the 22 farms evaluated. However, an analysis of the infective larvae showed that some drugs were effective against certain parasitic genera; i.e., levamisole was more effective against Haemonchus spp. and moxidectin against Trichostrongylus spp. In the additional FECRT performed at Farm # 1, moxidectin and nitroxynil were ineffective separately, but when applied in combination they were highly effective due to their efficacy against Haemonchus (nitroxynil) and Trichostrongylus (moxidectin), respectively. The use of the FECRT targeting the parasitic nematode species prevalent on farms may make it possible to choose more effective anthelmintics.


Resumo O objetivo deste trabalho foi apresentar dados sobre a eficácia de anti-helmínticos em rebanhos ovinos no Rio de Janeiro, Brasil, e discutir a interpretação do teste de redução da contagem de ovos nas fezes (TRCOF) para cada gênero de nematoide. A contagem de ovos fecais (OPG) e coprocultura pré e pós-tratamento foram realizadas para avaliar a eficácia e a prevalência geral do parasito, respectivamente. Um teste de eficácia adicional foi realizado na Fazenda # 1 após um ano do teste inicial. Resistência anti-helmíntica grave foi encontrada, não havendo sensibilidade no TRCOF em nenhuma das 22 fazendas avaliadas. No entanto, na análise das larvas infectantes observou-se que algumas drogas foram eficazes contra certos gêneros parasitários; por exemplo, o levamisol foi mais eficaz contra Haemonchus spp. e a moxidectina contra Trichostrongylus spp. No TRCOF adicional realizado na Fazenda 1, a moxidectina e o nitroxinil foram ineficazes separadamente, mas quando aplicados em combinação, foram altamente eficazes devido à sua eficácia contra Haemonchus spp. (nitroxinil) e Trichostrongylus spp. (moxidectina), respectivamente. O TRCOF visando às espécies de nematoides parasitas prevalentes nas fazendas pode possibilitar a escolha de anti-helmínticos mais eficazes.

ABSTRACT

The present study was conducted to detect the status of anthelmintic resistance of fenbendazole and morantel against gastrointestinal nematodes in goats of village Badhra, district Charkhi dadri, Haryana. Forty five goats with eggs per gram of more than or equal to 150 were divided into three groups i.e. G1, G2 and G3 of 15 animals each. Group G1 and G2 were treated with fenbendazole @ 10 mg/kg b.wt. orally and morantel @ 20 mg/ kg b.wt. orally, respectively. Group G3 served as untreated control. Faecal egg count of goats was ascertained on day of treatment (0 day) and 12th day post treatment (PT) of all groups by the modified McMaster technique. Pooled faecal cultures were made to recover infective larvae on day 0 and 12 PT. Results revealed that fenbendazole (G1) and morantel (G2) reduced the faecal egg counts by 76.47% and 70.80% on 12th day PT with upper and lower confidence levels as 89.30% and 48.23% and 82.43% and 51.48%, respectively indicating moderate anthelmintic resistance against both drugs. The post-treatment coproculture showed larvae of Haemonchus contortus and Strongyloides sp. Thus, the present study revealed presence of cross anthelmintic resistance against fenbendazole and morantel in goats of unorganized sector in Charkhi Dadri district of Haryana

The present study was conducted to detect the status of anthelmintic resistance of commonly used anthelmintic drugs viz. fenbendazole and morantel against gastrointestinal nematodes in sheep of village Badhra, district Charkhi dadri, Haryana. Forty five sheep with egg per gram of more than or equal to 150 were divided into three groups i.e. S1, S2 and S3 of 15 animals each. Group S1 and S2 were treated with fenbendazole @ 5 mg/kg b.wt. orally and morantel @ 10 mg/ kg b.wt. orally, respectively. Group S3 served as untreated control. Faecal samples were collected on the day of treatment (zero day) and 12th day post treatment (PT), from sheep of all groups and individual FEC were determined by the modified McMaster technique. Pooled faecal cultures were made to recover infective larvae on day 0 and 12 PT. Results revealed that fenbendazole (S1) reduced the FEC by 74.77% on 12th day PT with upper and lower confidence levels as 86.41% and 53.15%, respectively indicating moderate anthelmintic resistance. Morantel (S2) caused 49.84% reduction in FEC with upper and lower confidence levels as 65.04% and 28.04%, respectively indicating severe anthelmintic resistance. Thus, the present study revealed presence of cross anthelmintic resistance against fenbendazole and morantel in Haemonchus population in sheep of unorganized sector in Haryana as the post-treatment coproculture showed only Haemonchus contortus larvae

ABSTRACT

El monepantel es un nuevo antihelmíntico registrado en nuestro país exclusivamente para el control de los nematodes gastrointestinales de los ovinos y su uso ha sido direccionado mayormente hacia el control de parásitos resistentes a las clases de antihelmínticos disponibles actualmente. Estos mismos nematodes también parasitan a los caprinos, pero en estos rumiantes la patofisiología y la respuesta a los antihelmínticos es diferente, lo cual resulta en un mayor parasitismo y complejidad en el manejo de la resistencia. La presente comunicación informa sobre la eficacia del monepantel en dos hatos caprinos mantenidos bajo condiciones de campo y parasitados naturalmente por los géneros de nematodes gastrointestinales más comunes del área central de la Argentina (Haemonchus y Trichostrongylus) y portando alelos de resistencia múltiple (ivermectina y febendazole). El test de reducción en el conteo de huevos post tratamiento comparando diversas fórmulas, indicaron que en todos los hatos el monepantel por vía oral y a la dosis de 3,75 mg/kg de peso (1,5 veces mayor a la dosis ovina), resultó en eficacias del 99% al 100 %. Se realizan breves consideraciones sobre el uso potencial de esta droga en caprinos.


Monepantel is a new anthelmintic registered in Argentina exclusively for control of gastrointestinal nematodes of sheep and mostly directed toward controlling resistant parasites to current available classes of anthelmintics. The same nematodes also parasitize goats but pathophysiology and response are different in these hosts resulting in more severe parasitism and complexity in handling anthelmintic resistance. This report assess the efficacy of monepantel in goats maintained under field conditions and naturally parasitized by most common gastrointestinal nematodes from central Argentina (Haemonchus and Trichostrongylus) carrying alleles of multiple resistance (ivermectin and febendazole). According to the egg count reduction test, efficacies of 99% to 100% were observed after monepantel treatment at 3.75 mg/kg orally (1.5 ovine dose).

ABSTRACT

Abstract Frequent and inappropriate use of all classes of antiparasitic drugs in small ruminants has led to failures in their effectiveness, culminating in a global problem of anthelmintic resistance. Brazil stands out as one of the world’s leaders in publications about anthelmintic resistance, and for having the most numerous reports of this resistance in small ruminants in the Americas. These studies have involved mainly the fecal egg count reduction test (FECRT) and its correlation with field management practices. In vivoeffectiveness testing is conducted in areas where livestock is of greater economic significance, e.g., in the South (sheep) and Northeast (goats), or is important for research and economic centers, such as the Southeast (sheep). The most widely studied species is sheep, for which the widest range of drugs is also evaluated. Despite significant advances achieved in molecular research, laboratory analyses should include knowledge about the reality in the field so that they can become feasible for the producer. Moreover, molecular studies can be underpinned by the analysis of field studies, such as the maintenance of antiparasitic effectiveness over time and the mechanisms involved in this process.


Resumo O uso frequente e inapropriado de todas as classes de antiparasitários em pequenos ruminantes levou a falhas de eficácia, culminando na problemática global de resistência anti-helmíntica. O Brasil destaca-se como sendo um dos pioneiros nas publicações com resistência anti-helmíntica no mundo e por ter o maior número de relatos em pequenos ruminantes na América. Essas pesquisas envolvem principalmente o teste da redução da contagem de ovos nas fezes (TRCOF) e sua correlação com as práticas de manejo utilizadas no campo. Os estudos de testes de eficácia in vivo localizam-se em áreas onde há maior importância dos rebanhos como nas Regiões Sul (ovinos) e Nordeste (caprinos), ou com importância em polos de pesquisa e econômicos, como a região Sudeste (ovinos). Ovina é a espécie mais estudada e com maior gama de drogas avaliadas. Mesmo com grande avanço em pesquisas moleculares, as análises laboratoriais devem envolver o conhecimento da realidade do campo para que possam se tornar viáveis ao produtor. Além disso, a análise dos estudos de campo pode nortear estudos moleculares como, por exemplo, a manutenção da eficácia das drogas ao longo dos anos e os mecanismos envolvidos em tal processo.

ABSTRACT

A utilização de anti-helmínticos por longos períodos como principal medida de controle das parasitoses gastrintestinais de ruminantes levou a ineficácia aos levamisol, benzimidazóis e avermectinas. Este estudo descreve a atividade anti-helmíntica in vivo em populações naturais de nematoides trichostrongilídeos de caprinos. Foram selecionados 18 rebanhos provenientes dos biomas Caatinga (n=12) e Mata Atlântica (n=6), do Estado da Bahia, Brasil, criados em pastagens comunais em região semiárida. Grupos de oito a 10 animais foram tratados com albendazol (ABZ), ivermectina (IVM), levamisol (LEV), moxidectina (MOX) e closantel (CLOS). Os resultados do Teste de Redução da Contagem de Ovos nas Fezes indicaram resistência simultânea dos gêneros Haemonchus sp. e Trichostrongylus spp. para o ABZ, IVM, LEV, MOX e CLOS. As percentagens de eficácia variaram de 0-92%, 0-75%, 0-91%, 69-97% e 0-85% para o ABZ, IVM, LEV, MXD e CLOS, respectivamente, no bioma Caatinga e 0-59% para o ABZ e 9-59% para o IVM no bioma Mata Atlântica. Verificou-se nos rebanhos eficácia inferior a 95% para estes anti-helmínticos, com exceção de um único rebanho no qual a eficácia para MOX foi de 97%, o que sugere a presença de NGIs resistentes aos principais classes de anti-helmínticos em rebanhos caprinos destes biomas...


The use of anthelmintic drugs for long periods as the main measure control of gastrointestinal nematodes (GINs) has led to the inefficacy of levamisole, benzimidazoles and macrocyclic lactones. This study describes the in vivo anthelmintic activity against natural trichostrongyle nematodes populations in goats. We selected 18 herds from the Caatinga (n=12) and Mata Atlântica (n=6) biomes, Bahia State, Brazil, raised in communal pastures in semiarid region. Groups of 8 to 10 goats were treated with albendazole (ABZ), ivermectin (IVM), levamisole (LEV), moxidectin (MOX), and closantel (CLOS). The results of the Fecal Egg Count Reduction Test indicated simultaneous resistance of Haemonchus sp. and Trichostrongylus spp. genera against albendazole (ABZ), ivermectin (IVM), levamisole (LEV), moxidectin (MOX), and closantel (CLOS). The efficacy percentages ranged from 0 to 92%, 0 to 75%, 0 to 91%, 69 to 97%, and 0 to 85% for ABZ, IVM, LEV, MXD and CLOS respectively in the Caatinga bioma, and 0 to 59% for ABZ and 9 to 59% for IVM in the Mata Atlântica biome. Most herds showed efficacy lower than 95% for anthelmintics, with the exception of one herd in which the efficacy for MOX was 97%. ABSTRACT

Risk factors for anthelmintic resistance (AR) on bovine ranches were studied. Data were derived from a survey made to 50 ranch owners, who had conducted a faecal egg-count-reduction test. The questionnaire contained descriptors of bovine ranch management and nematode control. A case-control design study was undertaken and AR cases were present in 26 herds. Associations between the binary outcome variable (AR versus not AR) and risk factors recorded in the questionnaire were evaluated. Variables associated with the presence of AR at P< 0.15 and/or odds ratio (OR) > 2 were subjected to a multivariable logistic regression model. The main effects contributing to general AR (avermectin AVM and/or benzimidazole) in the final model were total number of annual treatments (OR 7.68; 95% CI 2.4 to 28.3) and use of more than 75% of AVM in the past (OR= 18.6; 95% CI 1.3 to 97.3), whereas for AVM resistance alone were total number of AVM annual treatments (OR= 11.5; 95% CI 2.9 to 45.5) and number of AVM Nov-Jan treatments (OR= 5.8; 95% CI 1.71 to 47.9). The results showed that treatment frequency, date of treatment and frequency of treatment in the past with a single drug were the main risk factors involved in AR development.


Fatores de risco para resistência anti-helmíntica (AR) em fazendas de criação de bovinos foram estudados de dados obtidos de um levantamento em 50 propriedades. Em todas foram conduzidos testes de redução de contagem de ovos (opg) e um questionário preenchido pelos proprietários sobre o manejo e o controle de verminose nessas fazendas. Um estudo com desenho de caso controlado foi realizado e casos de AR estavam presentes em 26 rebanhos. Associações foram avaliadas entre a variável binária produzida (AR versus sem AR) e fatores de risco registrados nos questionários. Variáveis associadas com o resultado de interesse a P<0.15 e/ou razão de prevalência (OR) > 2 foram usados num modelo de regressão logística multivariável. Os principais efeitos contribuintes para AR geral (avermectina AVM e/ou benzimidazole), no modelo final, foram número total de tratamentos anuais (OR 7,68; 95% IC 2,4 a 28,3) e uso no passado mais que 75% de AVM (OR= 18,6; 95% IC 1,3 a 97,3), e para resistência à AVM foram número total de tratamentos anuais (OR=11,5; 95% IC 2,9 a 45,5), número de tratamentos com AVM de novembro a janeiro (OR= 5,8; 95% IC 1,71 a 47,9). Estes resultados mostraram que a frequência dos tratamentos, a época do ano em que foram feitos os tratamentos e a frequência dos tratamentos no passado com uma única droga foram os principais fatores de risco implicado no desenvolvimento de AR.

ABSTRACT

A seleção e a crescente disseminação de nematoides resistentes aos anti-helmínticos mais comumente utilizados, benzimidazóis (BZs), imidazotiazóis e lactonas macrocíclicas (LMs), constituem um sério entrave na produção de pequenos ruminantes em todo o mundo. O uso de métodos eficientes e sensíveis para a detecção e o monitoramento da resistência anti-helmíntica no campo torna-se urgente, especialmente para os grupos de BZs e LMs, devido aos constantes relatos de resistência. A obtenção de um diagnóstico preciso e precoce da resistência é extremamente importante para auxiliar a tomada de decisão em programas de controle parasitário, com o objetivo de preservar a vida útil dos produtos e limitar o desenvolvimento da resistência nas populações de nematoides. Os testes in vivo e, mais recentemente, os testes in vitro têm sido desenvolvidos para a detecção de nematoides resistentes aos principais grupos de anti-helmínticos. No entanto, a disponibilidade de testes in vitro validados e o seu uso prático ainda são muito limitados. Embora o teste de redução na contagem de ovos nas fezes (TRCOF, in vivo - indireto) seja o principal método de escolha para a detecção de resistência no campo, vem recebendo críticas quanto à validade dos resultados, e passa por significativas modificações. Além disso, o desenvolvimento de técnicas moleculares a partir de alterações genômicas gerou avanços consideráveis nessa área de investigação, com o uso de mutações nos códons 167, 198 e 200 do gene da β-tubulina como principais SNPs (polimorfismos de nucleotídeo único; do inglês Single Nucleotide Polymorphisms) associados à resistência aos BZs. A presente revisão tem o objetivo de discutir os métodos de diagnóstico disponíveis para a detecção de resistência anti-helmíntica em nematoides de pequenos ruminantes, destacando progressos e obstáculos para seu uso na rotina laboratorial e no campo.


The selection and growing spread of resistant nematodes to the most commonly used anthelmintics, benzimidazoles (BZs), imidazoles and macrocyclic lactones (MLs), constitutes a serious obstacle of small ruminants production worldwide. The use of efficient and sensitive methods for detection and monitoring of anthelmintic resistance in the field becomes urgent, especially for the BZs and MLs groups due to its frequent resistant reports. Obtaining an early and accurate diagnosis of resistance is extremely important to aid decision-making regarding parasite control programs, with the objective to preserve the lifespan of existing products, and to limit the development of resistance in nematode populations. The in vivo tests and the more recent in vitro tests have been developed for the detection of nematode resistant to the major anthelmintic groups. However, the availability of validated in vitro tests and its practical use is still very limited. Although the faecal egg-count reduction test (FECRT, in vivo - indirect) is the primary method of choice for the detection of resistance in the field it has being criticized for its results and is receiving significant modifications. Moreover, the development of molecular techniques from genomic changes have generated considerable advances in this research area, with the use of mutations at codons 167, 198 and 200 of β-tubulin gene as the main SNPs (single nucleotide polymorphisms) associated with BZs resistance. This review aims to discuss the available diagnostic methods for the detection of anthelmintic resistance in nematodes of small ruminants, highlighting key developments and obstacles to its use in the laboratory and in the field.

Anthelmintic resistance is an increasing problem that threatens livestock production worldwide. Understanding of the genetic basis of benzimidazole resistance recently allowed the development of promising molecular diagnostic tools. In this study, isolates of Haemonchus contortus obtained from goats, sheep and buffaloes raised in Brazil were screened for presence of the polymorphism Phe200Tyr in the β-tubulin 1 gene, which confers resistance to benzimidazole. The allelic frequency of the mutation conferring resistance ranged from 7% to 43%, and indicated that resistance to benzimidazole could be found in nematodes isolated from all the ruminant species surveyed. Although significant variation in the frequency of the F200Y mutation was observed between different herds or host species, no significant variation could be found in populations isolated from animals within the same herd. These findings suggest that screening of samples from a few animals has the potential to provide information about the benzimidazole resistance status of the entire herd, which would enable a considerable reduction in the costs of diagnosis for the producer. Molecular diagnosis has practical advantages, since it can guide the choice of anthelmintic drug that will be used, before its application in the herd, thus reducing the economic losses driven by anthelmintic resistance.


A resistência aos anti-helmínticos é um problema crescente que ameaça a produção pecuária em todo o mundo. A compreensão da base genética da resistência ao benzimidazol permitiu, recentemente, o desenvolvimento de métodos diagnósticos moleculares promissores. Neste estudo, isolados de Haemonchus contortus obtidos a partir de rebanhos de caprinos, ovinos e bubalinos criados no Brasil foram avaliados quanto à presença do polimorfismo F200Y no gene da β-tubulina1, o qual confere resistência ao benzimidazol. A frequência alélica da mutação variou de 7% a 43%, indicando que a resistência ao benzimidazol pode ser encontrada em nematoides isolados a partir de todas as espécies de ruminantes pesquisadas. Embora tenha sido observada variação significativa das frequências de mutação F200Y entre rebanhos/espécies hospedeiros distintos, não foi encontrada variação significativa entre populações isoladas de animais dentro de um mesmo rebanho. Estes achados sugerem que a avaliação de amostras de alguns poucos animais tem o potencial de fornecer informações sobre o nível de resistência ao benzimidazol de todo o rebanho, possibilitando uma redução considerável dos custos de diagnóstico para o produtor. O diagnóstico molecular apresenta vantagens práticas, uma vez que pode guiar a escolha da base anti-helmíntica a ser utilizada antes da sua aplicação no rebanho, reduzindo, portanto, as perdas ocasionadas pela resistência aos fármacos anti-helmínticos.

ABSTRACT

Eighty-four half-blood Gir × Holstein (F1) calves aged six months who were naturally infected by gastrointestinal helminths and maintained in rotational grazing received different anthelmintic treatments. Group A received anthelmintics according to the usual management in the property (eight treatments, seven including a macrocyclic lactone agent). Group B received strategic treatment (ivermectin 3.15 percent) at the beginning and at the end of the rainy period. Eggs per gram of feces (EPG) counts and genus of larvae from fecal cultures were determined on a monthly basis from April 2002 to December 2003. There was no significant reduction (p > 0.05) in EPG counts in any group after anthelminthic treatment, and the larvae in fecal cultures observed were Cooperia, Haemonchus, Oesophagostomum and a few Trichostrongylus. Cooperia was the most prevalent genus in the first four months of the experiment and Haemonchus in the following months. In 2003, tracer calves were introduced onto the pastures monthly and they showed high nematode burden many times throughout the year, and Cooperia punctata and Haemonchus contortus were the main species identified. The results suggest that there is anthelminthic resistance in this farm, mainly to macrocyclic lactones, and the development of immunity by crossbred animals was vital to reduce nematode burden.


Oitenta e quatro bezerras meio sangue Gir × holandês (F1) com seis meses de idade, naturalmente infectadas por helmintos gastrintestinais e mantidas em pastejo rotacionado receberam diferentes tratamentos anti-helmínticos. O grupo A recebeu anti-helmínticos segundo manejo empregado na propriedade (oito tratamentos, sete com produtos à base de lactonas macrocíclicas). O grupo B recebeu tratamento estratégico (ivermectina 3,15 por cento) no inicio e final de período chuvoso. Mensalmente, no período de abril de 2002 a dezembro de 2003, foram realizadas contagens de ovos por grama de fezes (OPG) e coproculturas. Não houve redução significativa (p > 0,05) nas contagens de OPG em nenhum dos grupos após os tratamentos anti-helmínticos, e as larvas encontradas na coprocultura foram Cooperia, Haemonchus, Oesophagostomum e poucos Trichostrongylus, com predominância de Cooperia nos quatro meses iniciais e Haemonchus nos meses seguintes. No ano de 2003, bezerros traçadores foram alocados mensalmente nos pastos, apresentando altas cargas parasitárias na maioria dos meses do ano, sendo as principais espécies identificadas: Cooperia punctata e Haemonchus contortus. Os resultados indicam que os nematódeos da fazenda apresentam resistência anti-helmíntica, principalmente às lactonas macrocíclicas, e o desenvolvimento de imunidade foi primordial para reduzir a carga parasitária dos animais mestiços.

Background & objectives: Albendazole, a commonly used anthelminthic drug that targets the polymerization of α- and β-tubulin dimer is currently co-administered with the antifilarial drug, diethylcarbamazine citrate (DEC) in the ongoing Global Programme for Elimination of Lymphatic Filariasis (GPELF). The experience in veterinary field has shown that there can be a rapid development of resistance to this drug, which therefore, needs to be monitored regularly in GPELF. Hence, we investigated the nucleotide polymorphism in the albendazole-binding domain of the isotype 1 β-tubulin gene from several populations of Wuchereria bancrofti and developed an AS-PCR assay useful in screening for sensitive/resistance alleles among parasite populations and also evaluated its utility. Methods: For studying the polymorphism of isotype 1 β-tubulin gene, a 475 bp fragment spanning exon 5 and 6 of the gene was amplified and sequenced from the genomic DNA of W. bancrofti collected from six geographic regions of India. An allele specific (AS) PCR for screening albendazole sensitivity/resistance was developed and a total of 55 mf samples from blood smears on slides collected from Thiruvannamalai, Thanjavur and Puducherry were screened. Selective therapy with DEC was in place in three areas, mass drug administration (MDA) with DEC alone was implemented in four areas, while DEC plus albendazole was administered in one district. Results: The analysis of the nucleotide sequence of the fragment from 20 W. bancrofti populations showed the domain to be highly conserved. An allele-specific PCR assay developed was used to detect sensitive/ resistance alleles among 55 isolates of W. bancrofti and no albendazole resistance alleles were detected among the populations tested. Interpretation & conclusion: The drug-binding domain of isotype 1 β-tubulin gene of W. bancrofti from different geographical locations was highly conserved. The AS-PCR developed showed potential application as a tool for monitoring albendazole sensitivity/resistance alleles among W. bancrofti populations, in areas where combination therapy of DEC-albendazole is being mass administered in the LF elimination programme.
